Isolation and ITS Sequence Identification of the Pathogen Causing Red Rot Disease on Sugarcane

Abstract:

Five strains (CF-1, CF-2, CF-3, CF-4, CF-5) of pathogenic fungi were isolated and pathogen by the typical symptomatic tissues of sugarcane red rot with which single spore isolates, morphological observation, pathogenicity test, and ITS molecular sequence analysis were used to identify the pathogen. The results showed that both CF-1 and CF-4 could cause symptoms consistent with those observed in the fields. The amplification of the gene fragments of pathogenic bacteria by ITS1 and ITS4 showed that CF-1 showed 99.00% of identity with KU933924.1 and the strain CF-4 showed 99.42% of identity with MH854879.1. CF-1 was identified as Colletotrichum falcatum Went. and CF-4 as Nigrospora sphaerica (Sacc.) E.W. Mason. by culture. shape, morphological observation and ITS sequence analysis.
